As you appear to have a novice builder it is possible that the different hues are a result of different batches of plaster. One has to be careful about adding the same amount of sand, color, water, etc. as well as being consistent with the trowel.
Yes mold can grow on plaster I seen it grow on stucco and tile roofs. It can grow anywhere.
As to your photo, it appears to me that there is some type of damage, but I am not a mold expert and would so state "Apparent discoloration located in the upstairs xxxxxx room. Discoloration could be due to a variety of reasons including roof leak, water damage or possilbe organic infestion by source I am not qualified to determine. Lender/Client is advised of the visual inspection and appraiser suggest that if they have a concern that a qualified expert be employed by them to determine the reason behind the discoloration."
Standing back a few feet it looks "dirty" but getting up close the "dirt" is really tiny little dots about half the size of a pencil point. Some are black, some are brownish and some are kind of greenish. Could this be mold or fungus?
The "dots " description would be consistent with sand that has risen to the top of the plaster
